Flow cytometric analysis of Human TCR α/β expression on human PBMC (human peripheral blood mononuclear cells). Human PBMC were stained with PerCP-Cy5.5 Mouse IgM, κ Isotype Control (black line histogram) or SDT PerCP-Cy5.5 Mouse Anti- Human TCR α/β Antibody (red line histogram) at 1.25μl/test treated with True-Stain Monocyte Blocker™, cells without incubation with primary antibody and secondary antibody (blue line histogram) was used as unlabeled control. Total viable cells, as determined by Fixable Viability Dye 515 (S0D0013), were used for analysis. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.

Background
TCR α/β protein is the defining heterodimer of the αβ T-cell lineage, a trans-membrane clonotypic receptor whose extracellular N-terminal variable (V) and joining (J) domains of the α-chain pair with the V, diversity (D) and J domains of the β-chain to create a single antigen-binding site that is stabilized by the constant (C) domains and an inter-chain disulfide, while the short cytoplasmic tails lack intrinsic signaling capacity and therefore associate non-covalently with the CD3 γ, δ, ε and ζ dimers to form the intact T-cell receptor complex that triggers phosphorylation cascades upon recognition of a specific peptide bound to a self-MHC molecule, thereby initiating the adaptive immune response.
