Flow cytometric analysis of CD3d expression on human PBMCs (human peripheral blood mononuclear cells). Human PBMCs were stained with Brilliant Violet 421™ Mouse Anti-Human CD19 Antibody and either PE Mouse IgG2a Isotype Control (left panel) or SDT PE Mouse Anti-Human CD3d Antibody (right panel) at 5 μl/test. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.

Background
CD3d (CD3-delta) is a 19 kDa transmembrane glycoprotein that together with CD3ε, CD3γ and CD3ζ forms the CD3 signaling complex, which is tightly associated with the T-cell receptor (TCR) on thymocytes and mature T lymphocytes; upon antigen/MHC engagement the CD3 chains become phosphorylated on their cytoplasmic ITAMs by Lck and Fyn, initiating the cascade that leads to T-cell activation, proliferation and differentiation, while during thymic development CD3d is indispensable for correct assembly and surface expression of the pre-TCR/TCR complex and thus for positive selection of CD4⁺ and CD8⁺ T cells.
