Background

CD73, also known as ecto-5'-nucleotidase (encoded by NT5E), is a glycosylphosphatidylinositol (GPI)-anchored cell-surface enzyme that catalyzes the final step of extracellular ATP degradation, converting AMP to immunosuppressive adenosine, thereby orchestrating tumor immune evasion, promoting angiogenesis and metastasis, and shaping an anti-inflammatory microenvironment by dampening CD8 T-cell cytotoxicity and enhancing Treg or myeloid suppressive functions across multiple solid cancers.

Flow cytometric analysis of human CD73 expression on human PBMC (human peripheral blood mononuclear cells). Human PBMC were stained with Alexa Fluor® 647 Mouse Anti-Human CD3 Antibody and either PE/Cy7 Mouse IgG2b, κ Isotype Control (left panel) or SDT PE/Cy7 Mouse Anti-Human CD73 Antibody (right panel) at 1.25μl/test treated with True-Stain Monocyte Blocker™. Total viable cells, as determined by Fixable Viability Dye 452 (S0D0021), were used for analysis. Flow cytometry and data analysis were performed using Beckman CytoFlex S and FlowJo™ software.
