Background

CD117, also known as c-KIT or stem cell factor receptor, is a 145-kDa type III transmembrane receptor tyrosine kinase encoded by the KIT proto-oncogene on chromosome 4q12; upon binding its ligand stem cell factor (SCF) it dimerizes and autophosphorylates, triggering downstream RAS-MAPK, PI3K-AKT, and JAK-STAT cascades that orchestrate melanogenesis, hematopoiesis, mast-cell development, gametogenesis, and interstitial cell of Cajal function, and gain-of-function mutationsmost commonly in exons 9, 11, 13, or 17drive constitutive kinase activity underlying gastrointestinal stromal tumors (GIST), systemic mastocytosis, and subsets of acute myeloid leukemia, making CD117 a key diagnostic immunohistochemical marker and therapeutic target for tyrosine kinase inhibitors such as imatinib, sunitinib, and avapritinib.

Flow cytometric analysis of CD117 expression on Human PBMCs. Human PBMCs (peripheral blood mononuclear cells) were stained with Brilliant Violet 421™ Mouse Anti-Human CD34 and either PE-Cy7 Mouse IgG1, κ Isotype Control (left panel) or PE-Cy7 Mouse Anti-Human CD117 Antibody (right panel) at 5 μl/test treated with True-Stain Monocyte Blocker™. Flow cytometry and data analysis were performed using Agilent NovoCyte Quanteon and FlowJo™ software.
