Background

PDE4 (phosphodiesterase-4) is a cAMP-specific phosphodiesterase that hydrolyzes the second messenger cyclic adenosine monophosphate (cAMP) to 5'-AMP, thereby terminating cAMP signaling cascades involved in inflammation, memory, cardiac contractility and immune cell regulation; the enzyme family comprises four subtypes (PDE4A-D) that differ in N-terminal targeting domains and regulatory Upstream Conserved Regions (UCR1/2) controlling dimerization and PKA-mediated phosphorylation, and selective inhibition of PDE4—for example with roflumilast or apremilast—elevates intracellular cAMP, suppressing pro-inflammatory cytokines (TNF-α, IL-23) and increasing anti-inflammatory IL-10, making PDE4 inhibitors clinically useful in psoriasis, psoriatic arthritis, Behçet’s disease and severe COPD.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-PDE4 antibody was used at 1/100 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
