Background
PCDH10 (Protocadherin 10) is a member of the protocadherin family within the cadherin superfamily, functioning as a calcium-dependent homophilic cell adhesion molecule localized on the cell membrane. Structurally, it contains six extracellular cadherin domains, a transmembrane domain, and a cytoplasmic tail that differs from classical cadherins, and can generate multiple isoforms through alternative splicing. PCDH10 plays critical roles in the central nervous system, participating in neural circuit establishment and maintenance, synapse formation, and actin assembly; in the pathological context, PCDH10 is widely regarded as a tumor suppressor, and its expression silencing due to hypermethylation of its gene promoter is closely associated with the development, progression, and poor prognosis of various cancers (such as pancreatic cancer). Given its dual importance in the nervous system and oncology, PCDH10 has become a hotspot in research for targeted therapy of neurological diseases and as a non-invasive diagnostic and prognostic biomarker in tumors.
