Background

PDE4 is a member of the phosphodiesterase 4 family, belonging to the 3',5'-cyclic nucleotide-specific phosphodiesterases. Its primary function is to specifically hydrolyze the second messenger cyclic AMP (cAMP), converting it into inactive 5'-AMP, thereby terminating cAMP-mediated cell signaling pathways. The PDE4 family comprises four subtypes (PDE4A, PDE4B, PDE4C, PDE4D), each generating multiple variants through alternative splicing, with N-terminal targeting domains determining their subcellular localization and interactions with signaling complexes. As an important negative regulator, PDE4 controls local cAMP concentrations, thereby regulating downstream protein kinase A (PKA) and Epac signaling pathways, which in turn affect processes such as cell contraction, migration, proliferation, inflammatory responses, and learning and memory. Due to its high expression in immune and inflammatory cells, PDE4 has become an important therapeutic target for chronic inflammatory diseases, with selective inhibitors (e.g., apremilast, roflumilast) developed for the treatment of psoriasis, ankylosing spondylitis, and chronic obstructive pulmonary disease (COPD). Additionally, PDE4 dysfunction is associated with diseases such as depression, Alzheimer's disease, and asthma, and its inhibitors have shown potential value in neuroprotection and antidepressant effects.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human brain. Anti-pan PDE4 antibody was used at 1/250 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
