Flow cytometric analysis of Mouse CD226 expression on C57BL/6 mouse splenocytes. C57BL/6 mouse splenocytes were stained with APC Rat Anti-Mouse CD8α Antibody and either Pacific Blue Rat IgG2b, κ Isotype Control (Left panel) or SDT Pacific Blue Rat Anti-Mouse CD226 Antibody (Right panel) at 5μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
CD226, also known as DNAM-1, is a transmembrane glycoprotein in the immunoglobulin superfamily that functions as a key co-stimulatory receptor on T cells, NK cells, monocytes, and platelets, where it binds the nectin-family ligands CD155 and CD112 to trigger phosphorylation of its intracellular tyrosine and serine residues, thereby recruiting LFA-1 and activating Src, PI3K-AKT, VAV1-ERK, and PLCγ2 signaling cascades that enhance immune synapse formation, cytotoxic granule release, and production of IFN-γ and other cytokines, thus promoting antiviral and antitumor immunity; its expression correlates with improved outcomes in cancer immunotherapy, while its genetic polymorphisms or down-regulation are linked to autoimmune diseases and tumor immune evasion, making CD226 an emerging target for immunomodulatory therapies.
