C57BL/6 mouse BMDMs (bone marrow-derived macrophages) were fixed with 4% paraformaldehyde, permeabilized with 0.1% Tween. The cells were then stained with APC Rat Anti-Mouse F4/80 Antibody and either Pacific Blue Rat IgG2a, κ Isotype Control (left panel) or SDT Pacific Blue Rat Anti-Mouse CD107a Antibody (right panel) at 2 μl/test. Total viable cells, as determined by Fixable Viability Dye 545 (S0B88802), were used for analysis. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
CD107a (also known as lysosome-associated membrane protein-1, LAMP-1) is a heavily N- and O-glycosylated type I transmembrane glycoprotein (~120 kDa) that constitutes ~50% of lysosomal membrane proteins and is expressed constitutively in late endosomes/lysosomes but also transiently on the plasma membrane of degranulating cytotoxic T cells, NK cells, mast cells, and other activated cells, where it serves as a widely used functional marker of degranulation; it participates in lysosomal biogenesis, autophagy, cholesterol homeostasis, and the presentation of carbohydrate ligands to selectins, while its surface exposure correlates with cytotoxic activity and it has been implicated in cancer metastasis, adhesion of immune cells to vascular endothelium, and as a marker in lysosomal storage disorders.
