Flow cytometric analysis of Human XCR1 expression on Human peripheral blood Leukocytes. Human peripheral blood Leukocytes were stained with APC Mouse Anti-Human CD141 Antibody and either SDT Pacific Blue Rat IgG2a, κ Isotype Control (Left panel) or Pacific Blue Rat Anti-Human XCR1 Antibody (Right panel) at 5 μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
XCR1 is a conserved, 7-transmembrane G-protein-coupled receptor selectively expressed on CD8α⁺ and CD103⁺ classical dendritic cells; its exclusive ligand XCL1 is secreted by activated CD8⁺ T and NK cells, forming a chemotactic feedback loop that recruits antigen-loaded DCs into lymphoid tissue and optimizes cross-presentation, Th1 cytotoxicity and antiviral/antitumor immunity, making the XCR1–XCL1 axis an emerging target for cancer immunotherapy and vaccine adjuvant design.
