Background

CD3d (CD3-delta) is a 19 kDa transmembrane glycoprotein that together with CD3ε, CD3γ and CD3ζ forms the CD3 signaling complex, which is tightly associated with the T-cell receptor (TCR) on thymocytes and mature T lymphocytes; upon antigen/MHC engagement the CD3 chains become phosphorylated on their cytoplasmic ITAMs by Lck and Fyn, initiating the cascade that leads to T-cell activation, proliferation and differentiation, while during thymic development CD3d is indispensable for correct assembly and surface expression of the pre-TCR/TCR complex and thus for positive selection of CD4⁺ and CD8⁺ T cells.

Flow cytometric analysis of Human CD3d expression on human peripheral blood leukocytes. Human peripheral blood leukocytes were stained with Alexa Fluor® 647 Mouse Anti-Human CD19 antibody and either Pacific Blue Mouse IgG2a Isotype Control (Left panel) or SDT Pacific Blue Mouse Anti-Human CD3d Antibody (Right panel) at 5 μl/test. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.
