Background

P58IPK (also known as DNAJC3 or ERdj6) is an endoplasmic reticulum-resident DnaJ family co-chaperone that was initially discovered as a cytosolic inhibitor of the PKR kinase during influenza virus infection, and was later shown to be induced by endoplasmic reticulum stress and to participate in negative feedback regulation of the unfolded protein response (UPR) by inhibiting PERK activity. The protein consists of an N-terminal endoplasmic reticulum-targeting signal sequence, an N-terminal domain containing nine tetratricopeptide repeat (TPR) motifs, and a conserved C-terminal J domain. Its J domain is attached to the end of the elongated TPR domain via a flexible linker, with the HPD motif located at the protein's edge, approximately 100 Å away from the substrate-binding site. P58IPK possesses dual functions as both a molecular chaperone and a co-chaperone: the hydrophobic groove of its TPR domain can directly bind unfolded or misfolded protein substrates, and it then binds the endoplasmic reticulum Hsp70 homolog BiP through its J domain, stimulating BiP's ATPase activity and promoting substrate transfer to BiP.
