Background

P2RY12 (purinergic receptor P2Y, G-protein coupled, 12) is a 22 kDa Gαi-coupled seven-transmembrane receptor that is best known as the primary platelet receptor for ADP, where it triggers Rap1- and αIIbβ3 integrin-dependent aggregation and is the target of widely used antithrombotic drugs such as clopidogrel; beyond hemostasis, the same receptor is expressed on osteoclasts, microglia, vascular smooth-muscle cells and certain neurons, mediating ADP-evoked bone resorption, microglial chemotaxis and synaptic pruning, foam-cell formation in atherosclerosis, and neuroinflammatory responses that can either exacerbate or protect against neurodegeneration depending on context.
