IF shows positive staining in paraffin-embedded human tonsil. Anti-p27 (Alexa Fluor® 700 Conjugate) antibody was used at 1/50 dilution (magenta) and incubated overnight at 4°C. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IF staining protocol.

Background
p27 Kip1 is a member of Cip/Kip family of proteins, leading to cell cycle arrest in G1 phase. Loss or reduced expression is associated with poor outcome in carcinoma of breast, prostate, GI tract and lung. In pathology, it can be used for the identification of thyroid adenoma and thyroid follicular carcinoma.
