Background

Osteoprotegerin (OPG/TNFRSF11B/OCIF) is a member of the tumor necrosis factor receptor superfamily, secreted by osteoblasts as a homodimeric protein.Its core function is as a decoy receptor: 1) By binding with high affinity to RANKL, it competitively blocks RANKL from activating its functional receptor RANK, thereby strongly inhibiting osteoclast differentiation and activation while promoting their apoptosis, making it a key negative regulator of bone resorption. 2) It can also bind to TRAIL (TNFSF10), exerting an anti-apoptotic effect.Bone homeostasis directly depends on the local RANKL/OPG concentration ratio.As a central molecule in maintaining bone metabolic balance, abnormalities in its mechanism are closely associated with osteoporosis. Furthermore, studies suggest it plays important roles in regulating lymph node development and preventing vascular calcification, while its interaction with TRAIL may have implications in oncology and other fields.
