Usage Feeder-free culture method of human embryonic stem cells hES and induced pluripotent stem cells IPS (1 hour required for operation)
1. Thaw the matrix glue in an ice bath in a refrigerator at 4°C. overnight, and slowly blow the matrix glue 5 times with a pre-cooled gun tip to mix evenly. Be careful not to create bubbles. If bubbles are generated, the bubbles are removed by short centrifugation at low speed with a palm centrifuge.
2. Place the cell culture plate in an incubator for preheating.
3. Use a pre-cooled gun tip to divide the thawed matrix glue.
4. The matrix collagen solution was diluted in a ratio of 1:100 with 4°C. pre-cooled serum-free medium, and then the matrix gel dilution was spread out evenly and completely covered the culture plate. The recommended amount of matrix gel dilution used to modify the plates is 300 ul/cm2
5. The culture plate containing the modification solution was allowed to stand at room temperature for one hour.
6. Suck off the modified solution, and immediately seed a mixture of stem cells and culture medium (mTeSR1) on the culture plate. Be careful not to dry out the surface of the modified culture plate.
Description The basement membrane is a layer of stromal membrane on the basal surface of epithelial cells in animals. OrganoGel is a matrix gel formed from a basement membrane component extracted from mouse tumor tissue. The main component of that matrix glue are laminin, collagen IV, heparan sulfate proteoglycans (Kleinman et al. 1986). At the same time, the matrix gel also contains a variety of growth factors, such as epidermal growth factor EFG, platelet-derived growth factor PDGF, nerve growth factor NGF, basic fibroblast growth factor FGF-2, beta-transforming growth factor TGF-beta, and insulin-like growth factor ILGF (Vukicevic et al. 1992).

PRODUCT CHARACTERISTICS: This product is liquid at 4°C, but is gel when heated to 37°C. After the matrix glue has solidified, it is re-placed at 4°C overnight and the matrix glue can be liquefied again.

General Notes The matrix glue begins to solidify at temperatures above 10°C., so try to handle the matrix glue on ice. In order to avoid the effect of enzymes on organoids during organoid passage, organoids can be released from the matrix gel by slowly blowing the matrix gel directly with 4°C pre-cooled basal medium.
