Background
ORAI3 is a four-transmembrane store-operated Ca²⁺ channel protein that, upon association with the endoplasmic-reticulum Ca²⁺ sensor STIM1, forms highly Ca²⁺-selective CRAC channels mediating store-operated Ca²⁺ entry (SOCE) essential for T- and B-cell activation, endothelial and smooth-muscell function, and metabolic reprogramming; beyond SOCE, ORAI3 can heteromerize with ORAI1 to create arachidonic-acid-activated, store-independent channels, is transcriptionally up-regulated by TGF-β to drive the epithelial–mesenchymal transition gene Snai1, and in many cancers its over-expression promotes stemness, migration, chemoresistance and tumour growth via both Ca²⁺-dependent and Ca²⁺-independent signalling, making it a potential prognostic biomarker and therapeutic target.
