| Usage | 1. The tissue stored at-80 ℃ is slowly thawed at-20 ℃ half an hour in advance and then embedded in OCT. The fixed tissue is dehydrated with sucrose solution and embedded in OCT. The fresh tissue filter paper can be directly embedded in OCT after absorbing water. 2. Put an appropriate amount of dry ice into the ice cutting machine, drop a small amount of OCT on the sample tray, place the tissue cut surface upward on the sample tray, drop OCT embedding agent around the tissue to completely cover the tissue, and place the sample tray on the quick freezing table. Quickly freeze and embed it for about 10-20min. After the OCT turns white and hardens, you can slice it. 3. Fix the sample holder on the microtome. First, roughly trim the tissue surface and flatten it to the target observation site, and then start patching. Attach the tissue to the anti-detachment slide, mark the slide, and freeze the section. After drying, put it at-20 ℃/-80 ℃ for later use. 4. The sections were stained according to the requirements of subsequent experiments. |

| Description | OCT embedding agent is a water-soluble mixture of polyethylene glycol and polyvinyl alcohol, which has been widely used in the preparation of frozen tissue sections. After being embedded with OCT embedding agent, tissue can support tissue, increase tissue continuity, and reduce wrinkles and fragmentation. Because the OCT mixture is water-soluble, the sections dissolve after rinsing in water, and the background staining will not be increased in subsequent staining. It is an ideal medium for making frozen sections, so it is also called OCT frozen section embedding agent. |
| General Notes | 1. When adding OCT embedding agent dropwise, it should be uniform and try to avoid bubbles. 2. When embedding, pay attention to the position of the cut surface and the embedding direction of the sample. 3. The remaining OCT embedded blocks can be marked and stored in cryopreservation tubes at-80 ℃ for later use. 4. Fresh tissue should be OCT embedded within 30 minutes after tissue resection to make frozen sections. Tissues that cannot be treated in time can be stored at-80 ℃ after quick-frozen with liquid nitrogen or fixed with fixative and dehydrated with sucrose before OCT embedding. 5. Before using OCT embedding agent, please carefully check whether there are bubbles in the bottle. If there are bubbles, it needs to be stationary or centrifuged, and wait until the bubbles are eliminated before using. |
| Storage Temp. | Store at room temperature, shelf life is 2 years. |
