Background

Occludin is a approximately 65 kDa four-pass transmembrane protein, identified and named in 1993 as a pivotal component of tight junctions (TJs), primarily localized at the apical lateral membranes of epithelial and endothelial cells; featuring two extracellular loops, one intracellular loop, and a long C-terminal cytoplasmic tail, its extracellular loops mediate "zipper-like" sealing between adjacent cells via homo- or heterophilic interactions, thereby establishing a selective permeability barrier that strictly regulates paracellular transport and maintains cell polarity, while its C-terminus anchors tight junctions to the actin cytoskeleton by binding to cytoplasmic scaffolding proteins such as ZO-1, subsequently participating in the fine-tuned regulation of cell signaling, gene expression, proliferation, and differentiation, playing an indispensable central role in the construction of the blood-brain barrier, maintenance of tissue homeostasis, and pathological processes including inflammation and tumor metastasis.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
