Background
NTRK1/TrkA, encoded on chromosome 1q23.1, is a 140 kDa high-affinity receptor tyrosine kinase for nerve growth factor (NGF) that, upon ligand-induced dimerization and autophosphorylation at Tyr490, Tyr751 and Tyr785, activates the PI3K/Akt, Ras/MAPK and PLCγ pathways to orchestrate neuronal survival, differentiation, nociception and evasion of apoptosis, while its fusion proteins or mutations can drive oncogenesis and are targeted by TRK inhibitors.
