Background

NOX4, a constitutively active member of the NADPH oxidase family, is widely expressed in human tissues (including kidney, heart, liver, and brain) and localizes primarily to the outer mitochondrial membrane, where it catalyzes the reduction of molecular oxygen to hydrogen peroxide (H₂O₂) without requiring cytosolic regulatory subunits; it associates only with p22phox and can be further activated by Poldip2. Functionally, NOX4-generated reactive oxygen species (ROS) regulate diverse cellular processes such as proliferation, differentiation, apoptosis, senescence, and mitochondrial dysfunction, and it contributes to pathologies including cancer (e.g., breast and kidney cancer), fibrosis, atherosclerosis, and stroke. Recent studies also reveal NOX4's presence in the nucleus of kidney cancer cells, where its expression predicts resistance to targeted therapies and poor survival .
