Background

NOX2/gp91phox, encoded by the CYBB gene on chromosome X, is a 570-amino-acid, ~91 kDa glycosylated flavohemoprotein that forms the catalytic core of the phagocyte NADPH oxidase complex; its N-terminal six-transmembrane α-helical domain anchors it to the plasma membrane, binds p22phox, and ligates two non-identical hemes that sequentially transfer electrons across the membrane, while its C-terminal cytoplasmic tail docks NADPH and FAD, enabling a four-step redox cascade that reduces extracellular O₂ to microbicidal superoxide (O₂⁻), thereby driving the respiratory burst essential for killing invading pathogens; genetic loss-of-function mutations in gp91phox cause chronic granulomatous disease, whereas its overactivation contributes to oxidative stress implicated in atherosclerosis, hepatic fibrosis, and other inflammatory disorders.

IHC shows positive staining in paraffin-embedded human spleen. Anti-NOX2/gp91phox antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
