Background
NOX1 is a catalytic subunit of the NADPH oxidase family, with its core function being to catalyze the transfer of electrons from cytosolic NADPH to molecular oxygen via FAD and heme prosthetic groups, thereby generating superoxide anions and other reactive oxygen species. This protein features a typical six-transmembrane domain structure, with its C-terminus facing the cytosol and containing the catalytic domains that bind FAD and NADPH, while the N-terminal transmembrane region binds two hemes through conserved histidine residues, constituting a key link in the electron transport chain. Complete activation of NOX1 requires the formation of a stable complex with p22phox and is coordinately mediated by regulatory subunits such as NOXO1 (organizer) and NOXA1 (activator), while its activity is also dependent on the involvement of the small GTPase Rac1. This protein is expressed in various tissues including the colon, stomach, and vascular smooth muscle, and under physiological conditions it participates in processes such as host defense and cell proliferation; however, dysregulation of its activity or excessive production of reactive oxygen species is also closely associated with multiple pathological states, including atherosclerosis, cancer, and cardiovascular diseases.
