Background
Mouse Noggin is a 64 kDa secreted homodimeric glycoprotein that functions as a potent antagonist of Bone Morphogenetic Protein (BMP) signaling. Encoded by the Nog gene, Noggin directly binds BMP ligands (BMP-2, -4, -5, -7) and Growth Differentiation Factors (GDF-5, -6) with high affinity, preventing their interaction with cognate receptors and subsequent downstream Smad-dependent signaling cascade activation. The protein is expressed in key developmental organizers including the node, notochord, dorsal somite, and roof plate, where it establishes gradients of BMP activity essential for patterning.
In the adult mouse brain, Noggin is expressed in the subventricular zone (SVZ) where it creates a neurogenic niche by antagonizing endogenous BMPs. This promotes neuronal differentiation of neural stem cells while suppressing astrocytic fate, thereby maintaining neurogenesis throughout adulthood.
