Background
NINJ1, a 16 kDa evolutionary-conserved double-transmembrane protein first discovered as a nerve-injury-induced adhesion molecule, has emerged as the key executioner of plasma-membrane rupture (PMR) downstream of all major lytic cell-death pathways (pyroptosis, apoptosis, necroptosis, ferroptosis, PANoptosis, cuproptosis and parthanatos); upon death stimuli it oligomerizes into amphipathic filaments or “cookie-cutter” rings that breach the lipid bilayer, releasing damage-associated molecular patterns and propagating inflammation, while also regulating leukocyte trafficking, TLR4 signaling and blood–brain-barrier integrity, making it a promising therapeutic target in ischemia, sepsis, neuroinflammation and cancer.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-NINJ1 antibody was used at 1/1000 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
