Background

NF-κB2 p100 is a 100 kDa cytoplasmic precursor protein encoded by the nfkb2 gene that functions both as an IκB-like inhibitor of NF-κB—sequestering RelB and other subunits in the cytoplasm—and as the obligate source of the mature NF-κB2 p52 subunit generated via stimulus-induced, phosphorylation- and ubiquitin-dependent partial proteasomal processing triggered by the alternative NF-κB pathway kinases NIK and IKKα, thereby enabling p52-RelB dimers to translocate to the nucleus and drive transcriptional programs essential for lymphoid organogenesis, B-cell maturation and adaptive immunity.

Immunohistochemistry

IHC shows positive staining in paraffin-embedded human kidney. Anti- NF-κB2 p100/p52 antibody was used at 1/50 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry

ICC shows positive staining in HeLa cells. Anti- NF-κB2 p100/p52 antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
