Background
Nestin is a cytoskeletal component belonging to class VI intermediate filaments, initially identified as a marker of neuroepithelial stem cells and neural progenitor cells. It is structurally and expressively unique, featuring a short N-terminus and an extremely long C-terminal domain. Its expression is developmentally regulated: it is highly expressed in proliferating, undifferentiated progenitor cells and rapidly downregulated upon terminal differentiation, where it is largely replaced by other intermediate filament proteins (such as neurofilaments). Beyond the nervous system, Nestin is broadly expressed in local stem/progenitor cells in various tissues including skeletal muscle, heart, pancreas, liver, and hair follicles. As a cytoskeletal protein, Nestin typically co-polymerizes with other intermediate filament proteins (such as vimentin) to form heteropolymeric intermediate filament networks, participating in maintaining cell morphology, regulating cell migration, proliferation, and differentiation. Due to its aberrant high expression in various tumors (such as glioblastoma, melanoma, pancreatic cancer, and prostate cancer), which is closely associated with tumor malignancy, invasiveness, and poor prognosis, Nestin has been widely used as a reliable marker and a potential therapeutic target for cancer stem cells in solid tumors including gliomas.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded mouse E14 embryo brain. Anti-Nestin antibody was used at 1/1000 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. Counterstained with DAPI (Blue). Heat mediated antigen retrieval with EDTA buffer pH9.0 was performed before commencing with IF staining protocol.
