Background

MyoD1 (Myogenic Differentiation 1) is a basic helix-loop-helix (bHLH) transcription factor that serves as a master regulator of skeletal muscle differentiation, initiating the myogenic program by binding to E-box DNA sequences and activating the transcription of muscle-specific genes such as myogenin, MEF2, and muscle creatine kinase (MCK). Discovered in 1987, MyoD1 belongs to the MyoD family of myogenic regulatory factors (MRFs), which also includes Myf5, myogenin, and MRF4, and it can convert non-muscle cells, including fibroblasts, adipocytes, and even hepatocytes, into myoblasts through forced expression. The protein functions as a heterodimer with E-proteins (such as E12 and E47) to regulate cell cycle withdrawal and terminal differentiation, while its activity is modulated by various signaling pathways including p38 MAPK, PI3K/Akt, and Wnt signaling, as well as post-translational modifications like phosphorylation, acetylation, and ubiquitination. Beyond its canonical role in myogenesis, MyoD1 has been implicated in cancer biology, particularly in rhabdomyosarcoma where it serves as a diagnostic marker, and in metabolic regulation, highlighting its broad physiological significance.
