Background

The Murine Leukemia Virus (MuLV) p30 protein, also known as the CA (capsid) protein, is the major structural component of the MuLV viral core. As a critical product of the gag gene, it plays an essential role in viral assembly, forming the conical capsid that encases the viral RNA genome and enzymes. Due to its abundance and conservation, the p30 antigen serves as a definitive biomarker for the presence of the virus. Consequently, quantifying p30 levels via ELISA is a well-established and crucial method for determining the physical titer of MuLV particles. This measurement is vital in both virology research, for standardizing viral stocks, and in the biopharmaceutical industry, where it is used to monitor for retrovirus-like contaminants in biologics, such as vaccines and cell therapy products, ensuring patient safety.

Linearity
The concentrations of MuLV Titer p30 were measured and interpolated from the target standard curves and corrected for sample dilution.
The sample is undiluted samples are as follows: Virus supernatant was 4×10^4fold dilution. The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 7.12 ug/mL in human serum.

To assess the linearity of the assay, the sample was spiked with high concentrations of MuLV Titer p30 in various matrices and diluted with the appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay.
