Background

MST3, also known as Serine/Threonine Kinase 24 (STK24), is a member of the germinal center kinase III (GCKIII) subfamily within the Ste20-like kinase family and plays a pivotal role in regulating diverse cellular processes including apoptosis, cell migration, polarity, and autophagy. Structurally characterized by an N-terminal kinase domain and a C-terminal regulatory domain that facilitates homodimerization and interaction with scaffolding proteins such as STRIPAK components, MST3 functions primarily through phosphorylation of specific substrates like moesin and ezrin, thereby influencing cytoskeletal dynamics and membrane-cortex interactions. Its activity is tightly modulated by various signaling pathways, notably being activated under stress conditions or during immune responses, where it contributes to the regulation of T-cell activation and inflammatory signaling, while dysregulation of MST3 has been implicated in several pathological conditions, including cancer progression and neurodegenerative diseases, highlighting its significance as a potential therapeutic target in maintaining cellular homeostasis and signal transduction fidelity.
