Background

This four‑color panel is designed for detecting myeloid cell subsets, including monocytes, resident macrophages, granulocytes, and conventional dendritic cells in mouse tissue cells. CD11b was included as a general myeloid cell marker. Ly‑6C was included to distinguish monocytes from resident macrophages and granulocytes. Finally, CD11c and I-A/I-E (MHC class II) can identify conventional dendritic cells. The panel is compatible with most two‑laser flow cytometers. If desired, the panel can serve as a backbone panel, allowing for the addition of antibodies conjugated to various fluorophores to detect other markers of interest.

Mouse bone marrow were stained using the Mouse Myeloid Panel Kit consisting of anti-mouse CD11b FITC, anti-mouse Ly-6C PE, anti-mouse CD11c APC and anti-mouse I-A/I-E PerCP-Cy5.5 conjugated antibodies.
To analyze the data:
1. Create an FSC-A vs SSC-A dot plot and draw a gate around the leukocyte population.
2. Create a Fixable Viability Dye 777 (S0B88805) vs SSC-A dot plot with the leukocyte gate applied. Draw a gate around the live cell population.
3. Create a CD11b FITC-A vs SSC-A dot plot show the myeloid cells with the CD45+ gate applied.
4. Create a CD11b FITC-A vs Ly-6C PE-A dot plot with the CD45+ gate applied. Create gates to encompass CD11b+Ly-6C+ cells (monocytes), CD11b+Ly-6C- cells (Resident macrophages and granulocytes)
5. Create a CD11c APC-A vs I-A/I-E PerCP-Cy5.5-A dot plot with the CD11b+ gate applied. Create gates to encompass CD11c+I-A/I-E+ cells (conventional dendritic cell, cDC)

Mouse hepatocytes were stained using the Mouse Myeloid Panel Kit consisting of anti-mouse CD11b FITC, anti-mouse Ly-6C PE, anti-mouse CD11c APC and anti-mouse I-A/I-E PerCP-Cy5.5 conjugated antibodies.
To analyze the data:
1. Create an FSC-A vs SSC-A dot plot and draw a gate around the tissue cells excluding cellular debris.
2. Create a Fixable Viability Dye 777 (S0B88805) vs SSC-A dot plot with the tissue cells gate applied. Draw a gate around the live cell population.
3. Create a CD11b FITC-A vs SSC-A dot plot show the myeloid cells with the CD45+ gate applied.
4. Create a CD11b FITC-A vs Ly-6C PE-A dot plot with the CD45+ gate applied. Create gates to encompass CD11b+Ly-6C+ cells (monocytes), CD11b+Ly-6C- cells (Resident macrophages and granulocytes)
5. Create a CD11c APC-A vs I-A/I-E PerCP-Cy5.5-A dot plot with the CD11b+ gate applied. Create gates to encompass CD11c+I-A/I-E+ cells (conventional dendritic cell, cDC)

Mouse splenocytes were stained using the Mouse Myeloid Panel Kit consisting of anti-mouse CD11b FITC, anti-mouse Ly-6C PE, anti-mouse CD11c APC and anti-mouse I-A/I-E PerCP-Cy5.5 conjugated antibodies.
To analyze the data:
1. Create an FSC-A vs SSC-A dot plot and draw a gate around the lymphocyte population.
2. Create a Fixable Viability Dye 777 (S0B88805) vs SSC-A dot plot with the leukocyte gate applied. Draw a gate around the live cell population.
