Background

Mouse IL-12 is a critical pro-inflammatory cytokine that bridges innate and adaptive immunity. It is a heterodimeric protein composed of p35 (IL-12A) and p40 (IL-12B) subunits, forming the bioactive IL-12(p70). Primarily produced by antigen-presenting cells like dendritic cells and macrophages upon pathogenic stimulation, its pivotal role is to drive the differentiation of naïve CD4⁺ T cells into T helper 1 (Th1) cells and to potently induce interferon-gamma (IFN-γ) production from both T cells and natural killer (NK) cells. This establishes a robust cellular immune response essential for defense against intracellular pathogens such as viruses and bacteria. In biomedical research, Mouse IL-12 is a cornerstone for studying Th1-mediated immunity, evaluating vaccine efficacy, investigating antitumor immunotherapy strategies, and modeling autoimmune diseases like experimental autoimmune encephalomyelitis (EAE).

Linearity
The concentrations of Mouse IL-12 were measured and interpolated from the target standard curves and corrected for sample dilution.
#1 sample is undiluted samples are as follows: CD-1 Mouse serum. The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 14.72 pg/mL in CD-1 Mouse serum.
#2 sample is undiluted samples are as follows: BALB/c Mouse serum. The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 12.82 pg/mL in BALB/c Mouse serum.
#3 sample is undiluted samples are as follows: C57BL/6 Mouse serum. The interpolated dilution factor corrected values are plotted. The mean target concentration was determined to be 36.29 pg/mL in C57BL/6 Mouse serum.

To assess the linearity of the assay, sample was spiked with high concentrations of Mouse IL-12 P70 in various matrices and diluted with the appropriate Calibrator Diluent to produce samples with values within the dynamic range of the assay.
