Background

Mouse IFN-α1 (Interferon-alpha 1) is a type I interferon cytokine belonging to a multigene family that plays a critical role in the innate immune response against viral infections. This approximately 19-20 kDa protein is primarily produced by plasmacytoid dendritic cells (pDCs) and infected cells upon recognition of viral nucleic acids through pattern recognition receptors (PRRs) such as Toll-like receptors (TLR7/9) and RIG-I-like receptors. Upon secretion, IFN-α1 binds to the heterodimeric IFN-α/β receptor (IFNAR) complex composed of IFNAR1 and IFNAR2 subunits, triggering the JAK-STAT signaling pathway and inducing the expression of hundreds of interferon-stimulated genes (ISGs) that establish an "antiviral state" in neighboring cells. These ISGs encode proteins that inhibit viral replication at multiple stages, including protein kinase R (PKR), 2'-5'-oligoadenylate synthetase (OAS), and Mx GTPases. Beyond its antiviral functions, mouse IFN-α1 exhibits immunomodulatory properties by enhancing natural killer (NK) cell cytotoxicity, promoting dendritic cell maturation, and influencing T cell differentiation toward Th1 responses. The mouse IFN-α locus contains multiple subtypes (IFN-α1 through IFN-α13/14), with IFN-α1 being one of the most prominently expressed members during early viral infection. Structurally, it features a conserved four-helix bundle topology characteristic of the helical cytokine family, with species-specific variations that limit cross-reactivity between mouse and human proteins, necessitating the use of mouse-specific reagents for in vivo immunological studies.
