Background
Mouse C-X-C motif chemokine ligand 9 (CXCL9), also known as Monokine Induced by Interferon-Gamma (MIG), is a small cytokine belonging to the CXC chemokine family. It is a T-cell chemoattractant that is induced by interferon-gamma (IFN-γ). The protein functions as one of the three ligands for the chemokine receptor CXCR3, which is predominantly found on T cells. As an ELR-negative chemokine, CXCL9 selectively attracts activated T lymphocytes and plays a role in T-cell activation and angiogenesis. The mature, secreted mouse CXCL9 protein consists of approximately 105 amino acid residues with a molecular weight of around 12.2 kDa. It contains the four highly conserved cysteine residues characteristic of CXC chemokines and is commonly produced in E. coli expression systems for research purposes. Due to its role in Th1-mediated immune responses, CXCL9 is an important research target involved in conditions ranging from inflammatory diseases to cancer.

Spike Recovery
The recovery of Mouse CXCL9/MIG was evaluated in activated samples spiked with concentrations spanning the entire assay range.
Cross-reactivity
The heatmap displays OD450 ratios for serum samples from multiple species. Color intensity is proportional to the O.D.450 ratio (test species / reference species). Darker colors indicate higher cross-reactivity.
Spike-and-dilution Linearity
To evaluate assay linearity, two samples were spiked with high levels of Mouse CXCL9/MIG in different matrices and serially diluted with the corresponding Calibrator Diluent to fall within the assay's dynamic range.
Cross-reactivity
The cross-reactivity between Mouse CXCL9 and Human CXCL9 was assessed by testing both proteins in the same assay system. Serial dilutions of each protein were measured, and the dose-response curves were compared. No significant cross-reactivity was observed when Human CXCL9 was tested in the Mouse CXCL9 assay, as the signals remained within the background range of the assay.
Intra-Assay Precision (Precision within an assay)
Three samples of known concentration were tested fifteen times on one plate to assess intra-assay precision.
Inter-Assay Precision (Precision between assays)
Three samples of known concentration were tested in separate assays to assess inter-assay precision. Assays were performed with at least three lots of components.
Dilution Linearity
The concentrations of Mouse CXCL9/MIG were measured and interpolated from the target standard curves and corrected for sample dilution.
The leftmost bar in the graph shows biological samples diluted in Assay Diluent to 6.3% concentration. "undiluted" was defined as 6.3% biological sample. The subsequent bars represent two-fold serial dilutions of this reference. The mean target concentration was determined to be 883.6 pg/mL in Balb/c mouse EDTA plasma.
Dilution Linearity
The concentrations of Mouse CXCL9/MIG were measured and interpolated from the target standard curves and corrected for sample dilution.

The subsequent bars represent two-fold serial dilutions of this reference. The mean target concentration was determined to be 434.3 pg/mL in Balb/c mouse serum. The mean target concentration was determined to be 309.3 pg/mL in Balb/c mouse heparin plasma. The mean target concentration was determined to be 396.7 pg/mL in C57 mouse serum. The mean target concentration was determined to be 332.5 pg/mL in C57 mouse heparin plasma. The mean target concentration was determined to be 427.5 pg/mL in C57 mouse citrate plasma.
HOOK Effect Threshold
The upper limit of the HOOK effect was established at 100× the highest calibrator concentration (equivalent to a 2-log10 increase). No HOOK effect was observed below this threshold, confirming that samples within this concentration range yield reliable quantitative results without signal depression.
Determination of Minimum Detectable Dose (MDD)
The MDD was determined using three independent lots of assay components. For each lot, 19 replicate measurements of the diluent (zero calibrator) were performed. The mean (AVERAGE) and standard deviation (STDEV) of the 19 replicates were calculated. The MDD for each lot was then calculated according to the following formula:
MDD = 2 × STDEV + AVERAGE
