Background

CD96 (also known as TACTILE) is an immune checkpoint protein belonging to the immunoglobulin superfamily, closely related to TIGIT and CD226 (DNAM-1). It is primarily expressed on natural killer (NK) cells and T cells, where it interacts with its ligand, CD155 (PVR), and to a lesser extent, CD112 (PVRL2). CD96 functions as a co-inhibitory receptor, dampening immune responses by competing with the activating receptor CD226 for the same ligands.
