Flow cytometric analysis of Human PBMC (human peripheral blood mononuclear cells) labelling Human TIM-3 antibody at 1/2000 dilution (0.1 μg) / (Right panel) compared with a Mouse IgG1, κ Isotype Control / (Left panel). Goat Anti-Mouse IgG Alexa Fluor® 488 was used as the secondary antibody. Then cells were stained with CD56 - APC Antibody separately.

Background
TIM-3 (T-cell immunoglobulin and mucin-domain containing-3) is a transmembrane checkpoint receptor selectively expressed on interferon-γ-producing T cells, Tregs, and innate immune cells; engagement of its ligands (galectin-9, CEACAM1, HMGB1, phosphatidylserine) triggers inhibitory signaling that terminates Th1 responses, promotes T-cell exhaustion, and fosters tolerogenic dendritic cell activity, thereby serving as a key negative regulator of antitumor and antimicrobial immunity and representing an actionable target for cancer immunotherapy.
