Flow cytometric analysis of human peripheral blood lymphocytes labeled with anti-Human CD73 antibody at 1/200 dilution (1 μg) / (right panel) compared with a Mouse IgG1, κ Isotype Control / (left panel). Goat Anti- Mouse IgG Alexa Fluor® 488 was used as the secondary antibody. Then cells were stained with CD3 - Brilliant Violet 421™ Antibody separately. Flow cytometry and data analysis were performed using BD FACSymphony™ A1 and FlowJo™ software.

Background
CD73, also known as ecto-5'-nucleotidase (encoded by NT5E), is a glycosylphosphatidylinositol (GPI)-anchored cell-surface enzyme that catalyzes the final step of extracellular ATP degradation, converting AMP to immunosuppressive adenosine, thereby orchestrating tumor immune evasion, promoting angiogenesis and metastasis, and shaping an anti-inflammatory microenvironment by dampening CD8⁺ T-cell cytotoxicity and enhancing Treg or myeloid suppressive functions across multiple solid cancers.
