Background
CD54 is an 85–110 kDa transmembrane glycoprotein of the immunoglobulin superfamily that is expressed on endothelial, epithelial and immune cells; upon binding its ligands LFA-1 (CD11a/CD18) and Mac-1 (CD11b/CD18) it serves as the critical adhesion checkpoint that promotes firm leukocyte arrest on the vessel wall, enables subsequent diapedesis into inflamed tissue, and amplifies antigen-specific T-cell activation, thereby positioning it at the mechanistic center of inflammatory, autoimmune and anti-tumor immune responses.
