Background
CD32 (FcγRIIA) is a 40-kDa type I transmembrane glycoprotein of the Ig superfamily expressed on monocytes/macrophages, granulocytes, dendritic cells, B cells, platelets, and placental endothelium; it contains two extracellular Ig-like domains and functions as a low-affinity receptor for complexed/aggregated IgG (Kd < 10⁻⁷ M) that mediates phagocytosis, respiratory burst, ADCC, and immune complex clearance, with the FcγRIIA (CD32a) isoform delivering activating signals via an intracellular ITAM motif, while the FcγRIIB (CD32b) isoform delivers inhibitory signals via an ITIM motif, and polymorphisms and alternative splicing of the FCGR2A/FCGR2B/FCGR2C genes generate multiple isoforms with distinct expression patterns and functional roles .
