Background
B7-H4, also known as VTCN1 or B7x, is a ~50 kDa type-I trans-membrane glycoprotein of the B7 immunoglobulin superfamily that consists of an extracellular IgV-IgC domain pair, a single trans-membrane segment and a short cytoplasmic tail; it is expressed inducibly on antigen-presenting cells and constitutively on many epithelial cancers, where it acts as an immune-checkpoint ligand by binding an as-yet-unidentified receptor on activated CD4⁺ and CD8⁺ T cells, thereby arresting the cell cycle in G0/G1, suppressing IL-2 production and proliferation, and enabling tumours to evade immunity, making it an emerging target for cancer immunotherapy.
