Flow cytometric analysis of Foxp3 / Transcription Factor Staining Buffer Set fixed and permeabilized Human PBMC (human peripheral blood mononuclear cells) labelling Human Aiolos antibody at 1/200 dilution (1 μg) / (Right panel) compared with a Mouse IgG1, κ (Left panel) isotype control. Goat Anti - Mouse IgG Alexa Fluor® 647 was used as the secondary antibody. Then cells were stained with CD19 - Brilliant Violet 421™ antibody separately.

Background
Aiolos (encoded by IKZF3) is a zinc-finger transcription factor of the Ikaros family that is indispensable for lymphocyte biology. By recruiting chromatin-remodeling complexes to its target genes, it enforces quiescence in mature B cells, orchestrates immunoglobulin receptor editing, and cooperates with other Ikaros members to direct late-stage B-cell maturation. Its deletion or inactivating mutations unleash malignant transcriptional programs, making Aiolos a key tumor suppressor whose loss underpins B-cell acute lymphoblastic leukemia and multiple myeloma.
