Background

Mouse serum albumin (MSA) is a 66.5 kDa globular protein organized into three homologous α-helical domains (DI–DIII), each subdivided into two subdomains that create versatile binding pockets for transporting fatty acids, bilirubin, and drugs. Structurally, DIII contains the critical neonatal Fc receptor (FcRn) binding site, which mediates pH-dependent recycling and confers MSA's long half-life in murine circulation. In biomedical applications, MSA is extensively utilized as a blocking agent in immunoassays to reduce background noise, as a stabilizing supplement in cell culture to protect sensitive biomolecules from degradation, and as a biocompatible nanocarrier for targeted drug delivery through the enhanced permeability and retention (EPR) effect. Additionally, its high sequence homology with human serum albumin (≈80%) makes MSA an indispensable preclinical model for evaluating albumin-based therapeutics, pharmacokinetics, and drug conjugate stability in mouse disease models, thereby bridging fundamental protein chemistry with translational research.
