Background

Modified Trypsin, Mass Spectrometry Grade is produced through a multi-step process: the wild-type trypsin sequence is engineered to minimize autolysis, the modified enzyme is then expressed recombinantly to eliminate contaminating proteases, and finally subjected to reductive methylation to further enhance stability. The resulting product exhibits high purity and exceptional stability, making it ideal for a broad range of laboratory proteomics applications, including protein/peptide sequencing and identification, as well as HPLC peptide mapping.

Protocol

1 Dissolve Modified Trypsin by adding 20 to 200 μL of 2 mM HCL or 50 mM acetic acid, and prepare 2*Reaction buffer (100 mM Tris-HCl, 10 mM CaCl2, pH 8.0 @ 25°C)

2 Set-up a typical reaction as follows

2Incubate at 37°C for 2~4 h or overnight

Guidelines

1. Substrate must be in phosphate-free buffer to prevent calcium precipitation with both reconstituted enzyme and enzyme buffer.

2. Storage buffer:2 mM HCl or 50mM acetic acid
