Background
MMP3 (Matrix Metalloproteinase 3), also known as Stromelysin-1, is a key member of the matrix metalloproteinase family, renowned for its ability to degrade a wide range of extracellular matrix components—including proteoglycans, laminin, fibronectin, and type IV and type IX collagens—while also activating the pro-forms of other MMP family members (such as MMP1, MMP7, and MMP9), thereby serving as an upstream regulator in the tissue remodeling and matrix degradation cascade. Under normal physiological conditions, MMP3 expression is very low, but it can be rapidly induced by various inflammatory cytokines (such as IL-1β and TNF-α) and growth factors in response to tissue injury, inflammation, or the tumor microenvironment, and is primarily secreted by connective tissue cells, macrophages, and tumor stromal cells. This potent matrix-degrading capacity endows MMP3 with dual roles in multiple pathological processes: in joints, its overexpression is a key driver of cartilage destruction in osteoarthritis and rheumatoid arthritis; in the cardiovascular system, it participates in modulating atherosclerotic plaque instability; in tumor progression, MMP3 promotes tumor cell invasion and metastasis by degrading the basement membrane, while also potentially exerting tumor-suppressive effects in early stages by releasing matrix-bound pro-apoptotic factors. Due to its extensive involvement in disease processes, MMP3 is not only studied as a biomarker of disease activity but also represents a long-term goal for developing selective inhibitors to treat fibrosis, arthritis, and cancer.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human kidney. Anti-MMP3 antibody was used at 1/200 dilution, followed by a HRP Polymer for Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
Immunocytochemistry
ICC shows positive staining in U-87 MG cells. Anti-MMP3 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 100% ice-cold methanol and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).

Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
