Description
mIHC Dye (TSA540) is a high-performance fluorescent dye designed for multiplex immunohistochemistry (mIHC) based on tyramide signal amplification (TSA) technology. It enables robust, covalent fluorescent labeling on formalin-fixed paraffin-embedded (FFPE) tissues, frozen sections, and tissue microarray (TMA) slides, supporting multi-target, in situ biomarker detection in a single tissue section.
This reagent provides strong, stable fluorescence with Ex/Em: 520 nm / 540 nm, compatible with standard fluorescence microscopes and multispectral imaging systems. It delivers greatly amplified signals compared to conventional immunofluorescence, ideal for low-abundance antigens and spatial profiling of the tissue microenvironment.

Principle
TSA540 adopts horseradish peroxidase (HRP)-conjugated secondary antibodies to activate fluorescently labeled tyramide substrates. The activated tyramide rapidly forms covalent bonds with tyrosine residues near the antigen-antibody complex, forming stable fluorescent deposits. After that, the antibodies can be stripped by heat repair without removing the dye signal, allowing sequential labeling of multiple targets on the same tissue section to achieve multiplex staining.

Storage & Stability
Storage Condition: Store protected from light at 2–8 °C; avoid direct exposure to strong light.
Shelf Life: Stable for 12 months from the date of receipt when stored according to the required conditions.
