Background

MICB (MHC class I polypeptide-related sequence B) is a stress-inducible, heavily glycosylated transmembrane protein encoded by the MICB gene located on chromosome 6p21.33. It functions as a ligand for the NKG2D receptor, which is expressed on natural killer (NK) cells, CD8+ T cells, and γδ T cells. Upon binding, MICB activates the cytolytic response of these immune cells, promoting their ability to target and eliminate stressed or transformed cells, such as tumor cells or virus-infected cells. Unlike classical MHC class I molecules, MICB does not associate with β-2-microglobulin or bind peptides. Its expression is induced by cellular stress, including DNA damage and viral infections, making it a crucial component in both innate and adaptive immune responses. MICB is widely expressed in various tissues, including lymph nodes and bone marrow, highlighting its role in diverse physiological contexts.

Immunocytochemistry

ICC shows positive staining in K562 cells (top panel) and negative staining in THP-1 cells (below panel). Anti- MICB antibody was used at 1/100 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
