Background
The MFN1 protein, also known as Mitofusin 1, is a key regulator of mitochondrial fusion. It is a member of the dynamin-related GTPase family and is predominantly localized on the outer mitochondrial membrane. MFN1 plays a crucial role in maintaining mitochondrial morphology and function by mediating the fusion of mitochondrial outer membranes, which helps in the exchange of mitochondrial contents and the maintenance of mitochondrial DNA integrity. Dysregulation of MFN1 has been implicated in various diseases, including neurodegenerative disorders such as Parkinson's disease and Charcot-Marie-Tooth disease type 2A, where impaired mitochondrial dynamics contribute to cellular dysfunction and neuronal degeneration.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human kidney. Anti-MFN1 antibody was used at 1/200 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.
Immunocytochemistry
ICC shows positive staining in HeLa cells. Anti-MFN1 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
