Background

METTL1 is a member of the S-adenosylmethionine (SAM)-dependent methyltransferase family, primarily responsible for catalyzing the generation of N7-methylguanosine (m⁷G) modifications in tRNA. This protein typically forms a stable heterodimeric complex with WDR4, in which METTL1 serves as the catalytic subunit providing methyltransferase activity, while WDR4 acts as the auxiliary subunit responsible for substrate recognition and complex stability. At the functional level, METTL1-mediated tRNA m⁷G modification is essential for maintaining tRNA structural stability and regulating translation efficiency; this modification can influence the binding efficiency of tRNA to ribosomes by affecting its spatial conformation, thereby modulating global protein synthesis rates. Additionally, METTL1 is also involved in regulating the splicing of mRNA precursors containing m⁷G cap structures and plays important roles in various biological processes including cell cycle regulation, embryonic stem cell self-renewal, and DNA damage response. In terms of disease associations, METTL1 is upregulated in multiple tumors such as hepatocellular carcinoma, colorectal cancer, and lung cancer, where it promotes the translation of oncogenic mRNAs by enhancing tRNA m⁷G modification levels, thereby driving tumor cell proliferation and metastasis, making it a promising anticancer drug target. Furthermore, mutations or aberrant expression of METTL1 have also been linked to neurodevelopmental disorders such as intellectual disability and microcephaly.
