Background

MBD3 is a 32-kDa core subunit of the nuclear Mi-2/NuRD chromatin-remodeling complex that is essential for embryonic development in mice; it binds 5-methyl-cytosine DNA indirectly through a degenerate methyl-CpG-binding domain, couples ATP-dependent nucleosome sliding with histone deacetylase activity, and thereby orchestrates transcriptional repression, pluripotency exit, lineage specification, and heterochromatin formation across vertebrate genomes.

Immunocytochemistry

ICC shows positive staining in HeLa cells. Anti- MBD3 antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
