Background
MARCKS (Myristoylated Alanine-Rich C Kinase Substrate) is an evolutionarily conserved, intrinsically disordered peripheral membrane protein that serves as a key phosphorylation substrate for conventional and novel protein kinase C isoforms; by reversibly sequestering phosphatidylinositol-4,5-bisphosphate (PIP2) and cross-linking actin filaments, it integrates PKC, Ca2+/calmodulin, and small GTPase signaling to orchestrate dynamic membrane-cytoskeleton remodeling underlying cell adhesion, migration, endo/exocytosis, neurotransmitter release, and stem-cell differentiation, while its dysregulation contributes to cancer metastasis, inflammatory diseases, and neuronal disorders.

Immunohistochemistry
IHC shows positive staining in paraffin-embedded human cerebral cortex. Anti-MARCKS antibody was used at 1/500 dilution, followed by a HRP Polymer for Mouse & Rabbit IgG (ready to use). Counterstained with hematoxylin. Heat mediated antigen retrieval with Tris/EDTA buffer pH9.0 was performed before commencing with IHC staining protocol.

Immunocytochemistry
ICC shows positive staining in A431 cells (top panel) and negative staining in Jurkat cells (below panel). Anti-MARCKS antibody was used at 1/500 dilution (Green) and incubated overnight at 4°C. Goat polyclonal Antibody to Rabbit IgG - H&L (Alexa Fluor® 488) was used as secondary antibody at 1/1000 dilution. The cells were fixed with 4% PFA and permeabilized with 0.1% PBS-Triton X-100. Nuclei were counterstained with DAPI (Blue). Counterstain with tubulin (Red).
